I have a 2000 K3500 6.5 liter diesel, dooley cre cab and the manual has a section on 'power take-off'.

What is a power take-off and when would one use it? From reading the manual it seems quite involved and may hurt your drive train if done incorrectly.

Thanks in advance.

Hi John and Welcome to the DP forums. Power take off is usually used to run winches, dump boxes (hydralics), water pumps (Firetrucks) and other accessories with power diverted from the transmission.
It is an option and there is a fair amount of "extra" parts needed. If you have a standard trans there is a code on the top (paper) that tells a NewVenture dealer if you have all the correct parts inside the trans.
On a couple of our trucks there is a LOT of grinding of the gears if it is engaged incorrectly.
